【摘要】 目的 研究NFκB/p 50和Bcl2在大肠癌组织中的表达,分析二者在大肠癌发生、发展进程中的作用及意义。方法 运用免疫组化SP法检测NFκB/p 50和Bcl2在42例大肠癌及5例正常粘膜组织中的表达。结果 NFκB/p 50和Bcl2在大肠癌组织中的阳性表达率为61.9 %和52.4 %,明显高于正常粘膜,差异有统计学意义(P=0.008;P=0.026)。NFκB/p 50的表达与患者年龄、性别、肿块大小、Ducks分期、组织分化程度、淋巴结转移等临床病理参数均无统计学意义(P>0.05);Bcl2的表达与Dukes分期和淋巴结转移差异有统计学意义(P=0.031;P=0.002);大肠癌中NFκB/p 50和Bcl2的表达呈正相关(r=0.332,P=0.032)。结论 NFκB/p 50的高表达很可能促进了大肠癌的发生;NFκB/p 50可能通过调节Bcl2基因来抑制肿瘤细胞的凋亡。

【Abstract】 Objective To investigate the expressions of NFκB/p 50 and Bcl2 in carcinoma tissue of large intestine and their correlation.Methods The expressions of NFκB/p 50 and Bcl2 were detected by immunohistochemistry(SP) in 42 cases of carcinoma of large intestine and 5 cases of normal large intestine.Results The positive rates of NFκB/p 50 and Bcl2 expressions in carcinoma of large intestine were 61.9 % and 52.4 %.NFκB/p 50 and Bcl2 expression,which in carcinoma of large intestine group was higher than that in normal large intestine (P=0.008;P=0.026).The expressions of NFκB/p 50 was not statistically related with clinicpathological parameters of carcinoma of large intestine(P>0.05).The expressions of Bcl2 was statistically related with Dukes staging and lymphatic metastasis(P=0.031;P=0.002).Conclusion High expression of NFκB/p 50 probably promoted genesis of carcinoma of large intestine.NFκB/p 50 might inhibit tumour cell apoptosis by adjusting Bcl2.

大肠癌是消化系统常见的恶性肿瘤之一。大量研究表明,肿瘤的发生发展与细胞增殖、凋亡及基因异常有关。NFκB是迄今为止发现的功能最多的多向核转录因子,正常情况下,NFκB是以无活性的状态存在于细胞质中,在各种因素的作用下,NFκB被激活,调控其下游的凋亡抑制基因Bcl2或凋亡蛋白抑制因子家族成员Sorvirin[1]的活性,从而抑制细胞凋亡促进肿瘤的发生、发展。NFκB家族有多个亚单位,NFκB/p 50是其中一种研究较少而重要的亚单位。本研究采用免疫组织化学技术,检测大肠癌组织中NFκB/p 50和Bcl2蛋 [1] [2] [3] [4] 下一页 |
