【摘要】 ：［目的］寻找参麦注射液抗癌症恶病质的表达差异基因。［方法］建立结肠腺癌26荷瘤鼠癌症恶病质模型，随机分为两组后分别予等剂量的生理盐水、参麦注射液治疗。观察饮食量、体重、瘤体积变化等。第16d处死小鼠后取瘤。分别抽取各组瘤组织总RNA，应用Illiumina全基因组表达谱芯片（Mouse6 BeadChip），对差异表达基因进行分析探讨。［结果］参麦组恶病质小鼠去瘤体重明显重于空白对照组（P<0.01）；饮食量下降速度明显低于空白对照组（P<0.01）；与空白对照组对比，筛选得到参麦组差异表达基因共68条，其中56条呈上调趋势，12条下调，这些基因多数是小鼠肌cDNA文库中的基因，部分与代谢、免疫功能相关基因相关。［结论］参麦注射液有一定改善癌症恶病质的作用。其干预作用可能主要是通过调控小鼠肌肉组织以及与代谢、免疫功能等相关基因表达来实现的。

Abstract：［Objective］To detect the relevant differentially expressed genes of cancer cachexia(CC) induced by Shenmai injection in mice that carried colonic adenocarcinoma cells (colon26) with genechip technology. ［Methods］ Established colon26 CC mouse model, then randomly divided the mice into two groups and treated these mice with saline or Shenmai injection separately. Physiological conditions, body weight and tumor bulk were documented onca a day. To obtain the tumor tissue, all mice were executed on the 16th day. Total RNA extracted from the tumor tissue was submitted to Illumina Mouse6 BeadChip for whole genome gene expression profiling analysis. BeadStudio software was used to screen the differentially expressed genes between the two groups. ［Results］The decrease rate of diet quantity in the Shenmai group was significantly lower than that in the control group（P<0.01）, and the nontumor body weight of the Shenmai group was significantly heavier than that in the control group（P<0.01）. Sixtyeight differently expressed genes were identified between the Shemmai injection group and control group. Among these genes, fiftysix genes were upregulated and twelve genes were downregulated. Most of these genes belonged to the mouse muscle cDNA library and were related with immunity and metabolism of tumor. ［Conclus [1] [2] [3] [4] 下一页 |
