温度和培养基在申克孢子丝菌酵母相转化中的作用研究
BHI, 50 mg/kg goat blood BHI and 5 mg/kg GSBHI, and 5 plates of each media were prepared because isolates of each meida were incubated at 25 ℃, 35 ℃, 36 ℃, 37 ℃, and 38 ℃. Growth of Sporothrix schenckii was observed and rates of transformation were calculated. 【Results】 At 38 ℃, the transformations rates were 25%(the only one Strain converted yeast). At 35 ℃ and 37 ℃, the transformations rates were 60% and 85%, respectively. At 36 ℃, the transformations rates were 45%. There were 13 of 20 plates converting yeast when 5% goat blood BHI and 0.5%GSBHI were used, 9 of 20 plates when BHI was used, 4 of 20 plates when SDA and PDA were used. At 37 ℃, transformation rates of 3 isolates reached 90% on medium of BHI, 50 mg/kg goat blood BHI和5 mg/kg GSBHI; However, the transformation rates of 1 isolate were less than 10% on SDA, PDA, and BHI. 【Conclusion】 Temperature is the key factor that affects mycelium-to-yeast transformation of Sporothrix schenckii, nourishments of media may also be necessa
